Acute myeloid leukemia (AML) often develops after a "preleukemic" phase where mutated hematopoietic stem cells expand due to a fitness advantage. We use prospective models and patient samples to investigate how the duration of preleukemia and the burden of preleukemic clones influence the progression to AML. Our findings aim to deepen understanding of AML pathogenesis and inform clinical management of preleukemic conditions like clonal hematopoiesis.
